Filter photometer: Design and mode of operation

A filter photometer is an optical measuring instrument for routine analysis used to determine the concentration of substances in a solution. The construction of the instrument consists of a light source, a monochromator and a detector.

The light source generates radiation with a broad spectrum, which is passed through the monochromator. The monochromator separates the light spectrum into individual wavelengths and allows only one specific wavelength to pass through. This monochromatic light then hits the sample. Depending on the concentration of the substance to be measured, it absorbs part of the incident light.

The remaining intensity of the transmitted monochromatic beam is measured by the detector and output as an electronic signal. To determine the concentration of the substance, a calibration curve must first be created: Here, one measures the ratio between absorbed and irradiated intensity (absorption behavior) at various known concentrations. From this curve, it is then possible to read off the concentration of each unknown sample.

Filter photometers are very precise measuring instruments with high sensitivity to the slightest changes in the absorption behavior of samples - therefore they are often used in laboratory or industrial environments for quality control or analysis of various materials such as water samples or food ingredients.

Calibrating a filter photometer: this is how it's done

To calibrate a filter photometer, a few steps are necessary. First, make sure that the instrument is in a clean and stable condition. This includes cleaning the measuring cell as well as the filter.

  • Next, a reference sample must be measured. This is a solution with a known concentration of the analyte that is to be measured later. The measured values of this sample serve as the basis for the calibration curve.
  • Several standards containing different concentrations of the analyte should then be prepared. These can then be measured one after the other and, together with the reference sample, form the basis for the calibration curve.
  • The curve obtained can now be used to analyze unknown samples and determine their concentration.
  • When calibrating a filter photometer, it is also important to regularly check for accuracy and reproducibility by performing control measurements of standard solutions or interlaboratory tests with other laboratories.

These careful measures ensure that the result of an analysis is as accurate as possible and can therefore provide meaningful data.
